Overview

Uperio 200mg Tablet is a combination medication containing two active ingredients: sacubitril & valsartan. It is used for the treatment of heart failure with reduced ejection fraction (HFrEF). Sacubitril is a neprilysin inhibitor, while valsartan is an angiotensin II receptor blocker (ARB). This combination medication works by increasing levels of natriuretic peptides and inhibiting the renin-angiotensin-aldosterone system (RAAS), resulting in vasodilation and reduction of blood pressure.

Active Ingredients

  • Sacubitril: Sacubitril is a neprilysin inhibitor that inhibits the enzyme responsible for breaking down natriuretic peptides, such as atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP). By inhibiting neprilysin, sacubitril increases levels of these peptides, which promote vasodilation, diuresis, and natriuresis.
  • Valsartan: Valsartan is an angiotensin II receptor blocker (ARB) that blocks the action of angiotensin II on its receptors, leading to vasodilation, reduced aldosterone secretion, and decreased sodium and water retention.

Mechanism of Action

Uperio 200mg Tablet works through two complementary mechanisms:

  1. Sacubitril increases levels of natriuretic peptides, which promote vasodilation, diuresis, and natriuresis, thereby reducing blood pressure and preload on the heart.
  2. Valsartan blocks the vasoconstrictor and aldosterone-secreting effects of angiotensin II, leading to further vasodilation and reduction of blood pressure.

Uses

Uperio 200mg Tablet is indicated for the treatment of heart failure with reduced ejection fraction (HFrEF) to reduce the risk of cardiovascular death and hospitalization for heart failure.

Dosage and Administration

The recommended dosage of Uperio 200mg Tablet is one tablet taken orally twice daily. The dosage may be titrated based on individual patient response and tolerability.

Side Effects

Common side effects of Uperio 200mg Tablet may include:

  • Hypotension (low blood pressure)
  • Hyperkalemia (high potassium levels)
  • Renal impairment
  • Dizziness
  • Fatigue
  • Cough
  • Angioedema

Precautions

  • Uperio 200mg Tablet should not be used in patients with a history of angioedema related to previous angiotensin-converting enzyme (ACE) inhibitor or angiotensin II receptor blocker (ARB) therapy.
  • Patients should be monitored for hypotension, hyperkalemia, and renal function during treatment with Uperio.
  • Concomitant use of Uperio with other potassium-sparing diuretics, potassium supplements, or medications that increase potassium levels should be avoided.
